Detailed description of the DNA ordering
procedure
After login to LAREU, you
will see a pull-down menu on the right side
of “Change”, labeled “DNA”. Moving the mouse over
this menu point will give access to the ordering
procedure for the DNA-typing of your animals. In order to make the DNA
typing efficient, you are advised to first type the parents and then the
offsping. Please note that parentage verification can only be done when the
parents
are already DNA-typed within the LAREU system.
How to select the laboratory for the
DNA typing First of all you will select
one of the Laboratories working together with LAREU.
For this move the mouse over “DNA” and click on
fly-out “Select DNA laboratory”. You
have a choice between four laboratories, in Germany
(Certagen GmbH), France (Progenès), the Netherlands
(van Haeringen BV) and Belgium (van Haeringen polygen bvba) . For each laboratory you will
find some general information as well as the Terms
& Conditions with the special LAREU prices for the
services offered. Once a laboratory is selected
by pushing the “Select this lab” button, it will
stay your laboratory of choice, unless you change
it.
How to collect the DNA material from
your animal
After thorough tests, LAREU
and the DNA labs have decided on a simple method
for collecting the DNA material from your animals,
which you can apply yourself and does not require
a veterinarian. The preferred method is to use a
“Q tip”-like cotton stick, named Performagene,
produced by DNA-Genotek in Canada. We will call
this product “swab” in the following. The swab comes
on top of a screw cap of a plastic tube which contains a special liquid (see picture).
For collecting the DNA material the swab is introduced into the
nose of the animal, some 2.5-4 cm deep, and rubbed
gently against the nasal wall. Then, holding the tube in upright position not to spill any of the liquid
the swab is unscrewed, placed into a plastic tube in the liquid, and screwed tight.

How to order the Swabs
The swabs can be ordered using an order form
which is sent to you together with the confirmation
email, once you have ordered DNA for your animal
(see below). Print out the pdf, fill the desired
number of swabs, sign the form and send it by fax
to the number given on the order form, or mail it
to the laboratory address given.
How to order the DNA typing for your animal
The DNA typing of your animal proceeds in four
steps:
1. Logon to your user area at LAREU
and press “DNA”. Select the pull-down menu “order
DNA typing”. A list of your animals not yet DNA-typed
will appear. You can order this list by clicking
on one of the column names, e.g. “Animal name” to
get the animals ordered in alphabetic order. Press
the “Place Order”-button on the left side for the
animal you want to DNA-type. A new page will appear
with the data of the animal you selected (on this
page also the lab you have selected will appear).
On the line of the animal name the box “DNA typing”
is already ticked for you. You only have to accept
the “Terms & Conditions” of the selected laboratory
by ticking off the corresponding box (you can read
them again by clicking on the link). Then you press
“Order”. Clicking the “Order” button will automatically
send an email with the animal data and your address
to the laboratory. At the same time you will receive
an email from LAREU with the confirmation of the
order.
2. The confirmation email contains
two pdf-files appended, which you should print out.
One of the pdfs contains the DNA order form with
the animal information, the address of the laboratory,
and your address. The order is in two languages:
English, and German for Certagen, French for Progenès,
and Dutch for Van Haeringen BV. The second page
contains an order form for the swabs. If you do
not have any swabs yet, then fill the swab order
form, enter the number of desired swabs, sign it
and send it by fax or mail. If you have the swabs
already, proceed with point nr. 4.
3. If
you have ordered the swabs, they should arrive within
roughly a week. Please make sure that you have received
the correct swabs. In case you did not receive the
correct swabs, please notify LAREU (email to info@lareu.org).
4. Collect the DNA material from your animal
with the swabs as described above . Note down the
serial number of the swab together with the name
of your animal for later identification. On the
order form you note down the date of the DNA sampling,
the number of the swab, and the date of the submission.
Finally you sign the form. With your signature you authorize
the DNA lab to upload the DNA data to the LAREU database.
The second pdf contains
a bar code together with the name of your animal
and its LAREU number, which you cut out and glue
onto the swab tube. Order form and swab tube are
then sent to the laboratory.
5. When the
laboratory receives your order form, it will send
an invoice to your address as given in your LAREU
personal area. After your payment is received, the
DNA typing is carried out and the DNA data are uploaded
to the LAREU data base. When this happens, you will
receive an email from LAREU stating that you can
now look at the DNA data of your animal. The DNA
data established by the laboratory can be found
at the bottom of the detailed view of your animal.
The full DNA information is given by three lines,
where the allele lengths for each marker are displayed.
The LAREU system also generates a DNA number, which
will appear in the detailed view as well. The LAREU
DNA number has the following format: Lyyll000xxx,
where yy notes the year of the test, “ll” the laboratory
number, and “xxx” the internal order number within
the LAREU system. A typical number will look like
this: L122300278.
How is the parent verification
done
When the parents of your animal
have been DNA-typed (use the same procedure as above),
you can order a verification of the parentage, done
by the laboratory. For this you select your animal
from the list of DNA-typed animals (select “Order
DNA typing” and select animals “with DNA typing”).
Select “Place Order”,
and tick off the boxes for “mother” and /
or “father” (please note that these boxes can only be ticked
if the DNA typing of the corresponding parent has been done in the
LAREU system). Then click on the “order” button. You will
receive a confirmation email with the two pdf files appended. Print out
the order form, fill it out and sign it. Send it to the lab together
with the swab. In case you only need parent verification (cria is already
DNA-typed), only the order form is sent.
As in the case above you will receive an
invoice with the cost. After payment the parentage
verification (and the DNA typing of the cria, if requested) is done
and you will receive an email
with the confirmation.
As proof of the correct verification
of the parents, the LAREU DNA number will receive
an appendix with “FV” if the father was verified,
and “MV”, if the mother was verified. A typical
DNA number with both parents verified will look
like this: L1223000278FVMV.
In case the parentage
could not be confirmed, this will be stated in the
confirmation email and no appendix to the LAREU
DNA number will be generated.
In the case the parentae could not be verified, the confirmation email
will contain a corresponding comment. In addition, the appendix to the
LAREU DNA number will not be set.

This is the official set of DNA markers, defined
recently by the International Society of Animal Genetics
(ISAG). This set will be used for DNA testing by the
DNA laboratories working together with LAREU. These
laboratories are certified according to the ISO 17025
standard.
